摘要 用微波辅助法提取灵芝子实体中的多糖,优化灵芝子实体中多糖的提取工艺及考察多糖的抗氧化能力。依据单因素实验结果确定料液比、提取时间、提取温度、提取功率为自变量,多糖提取率为响应值,设计响应面实验方案。最佳提取工艺条件为:料液比1:20(g/mL)、提取时间20 min、提取温度80℃、提取功率570 W。在该工艺条件下进行3次验证实验,多糖平均提取率为1.15%;灵芝多糖抗氧化实验的DPPH自由基清除率的IC50值为0.40 mg/mL,ABTS自由基清除率的IC50值为0.21 mg/mL,总还原力能力较好。微波提取灵芝多糖提取率高,抗氧化能力好,为灵芝多糖的进一步开发提供理论基础。

Abstract: Microwave assisted extraction method was used to extract polysaccharides from the fruited bodies of Ganoderma lucidum. The extraction process of polysaccharides was optimized and the antioxidant activity was investigated. According to the single factor experimental results, the solid-liquid ratio, extraction time, extraction temperature and extraction power were determined as independent variables, and the content of polysaccharide was chosen as the response value. The optimal extraction conditions were as follows: solid-liquid ratio 1:20(g/mL), extraction time 20 min, extraction temperature 80℃ and extraction power 570 W. The average polysaccharide content was 1.15% by three verification experiments under this process condition. The IC50 value of DPPH free radical scavenging rate and the IC50 value of ABTS free radical scavenging rate were 0.40 mg/mL and 0.21 mg/mL respectively, and the total reducing power of polysaccharide was better. The polysaccharide has antioxidant ability, which provides theoretical basis for further development of Ganoderma lucidum polysaccharide.
