基于网络药理学的龙胆泻肝汤肝毒性机制研究

摘要 探讨了龙胆泻肝汤潜在作用靶点、与肝毒性交集靶点、蛋白质互作网络、GO分析以及KEGG分析。通过TCMSP搜索龙胆泻肝汤的成分,并使用SwissTargetPrediction数据库寻找潜在作用靶点,发现了共有881个潜在作用靶点。利用GeneCards数据库搜索人类相关肝毒性靶点,并与龙胆泻肝汤的成分相关靶点数据取交集,得到178个共同靶点。通过STRING数据库构建蛋白质互作网络,最终确定了27个核心靶点。利用核心靶点进行GO分析,结果显示这些靶点与多个细胞组分、生物过程和分子功能都存在相关。KEGG富集分析结果显示与肝损伤相关的通路主要包括癌症途径、丙型肝炎、肝细胞癌等。本研究结果为龙胆泻肝汤的药理作用和潜在毒性提供了一定的参考。

Abstract: Potential targets of action, targets of intersection with hepatotoxicity, protein interaction network, GO analysis, and KEGG analysis of Longdan Xiegan Decoction (LXD) were investigated. A total of 881 potential targets of action were identified by searching the ingredients of LXD through TCMSP and using the SwissTargetPrediction database for potential targets of action. The GeneCards database was used to search for human-related hepatotoxicity targets and intersected with the data of targets related to the ingredients of LXD to obtain 178 common targets. Protein interaction networks were constructed through the STRING database, and 27 core targets were finally identified. Further GO analyses showed that these targets were involved in multiple cellular components, biological processes and molecular functions. The results of KEGG enrichment analysis showed that the pathways associated with liver injury mainly included cancer pathway, hepatitis C, and hepatocellular carcinoma. The results of this study provide some reference for the pharmacological effects and potential toxicity of LXD.
