参玫五味颗粒剂制备工艺及体外活性研究

摘要 探索参玫五味颗粒剂的制备工艺及其体外活性。采用单因素及正交实验,对参玫五味总皂苷提取工艺及颗粒剂制备工艺进行优化;利用紫外-可见分光光度法测定参玫五味总皂苷含量;通过测定α-葡萄糖苷酶活性的抑制率及DPPH自由基清除率考察了总皂苷的体外活性。参玫五味总皂苷的最佳提取工艺为:提取溶剂为70%乙醇溶液,料液比为1:10(g/mL),80 ℃提取时间2次,每次2.5 h,在此条件下,参玫五味总皂苷的提取率为7.36 mg/g;参玫五味颗粒剂最佳制备工艺为:浸膏粉与稀释剂质量比为1:1(g:g),稀释剂中乳糖与糊精的质量比为2:1(g:g),三氯蔗糖为矫味剂,润湿剂为85%乙醇溶液。体外活性试验结果表明,参玫五味总皂苷具有较好的体外降血糖及抗氧化活性。为参玫五味颗粒剂在保健食品方向的进一步开发提供了依据。

Abstract: The extraction and preparation of total saponins from Shenmei Wuwei granules were studied, and the activity in vitro of the extract was explored.Single factor and orthogonal experiment were applied to optimize the extraction technology of total saponins from Shenmei Wuwei materials and the preparation technology of granules.The content of total saponins in extract was determined by ultraviolet spectrophotometry, and the activity in vitro of the total saponins extracted from Shenmei Wuwei granules was discovered by α-glucosidase inhibition rate and DPPH free radical scavenging. The optimum extraction technology was obtained as follows: extraction solvent, temperature, extraction time, material-to-liquid ratio and extract times were 70% ethanol solution, 80 ℃, 1:10 (g/mL), 2.5 h and 2 times respectively which offered the extraction rate of total saponins from Shenmei Wuwei materials to 7.36 mg/g. The preparation process of Shenmei Wuwei granules was as follows: the mass ratio of extract powder to diluent was 1:1 (g:g), the mass ratio of lactose to dextrin in diluent was 2:1 (g:g), sucralose was used as the flavor correcting agent, and the wetting agent was 85%. ethanol solution. Activity test results showed that the total saponins of Shenmei Wuwei had good hypoglycemic and antioxidant activities in vitro. Shenmei Wuwei granules provides basis for its further research and development in health food field.
