The direct extraction and preparation method of extracting ginseng extract which is rich in Rg3 and Rg5 from ginseng root powder was studied in the paper, and the technical process optimization was carried out by single factor experiment and orthogonal experiment. In the study, ginseng root powder was used as raw material, natural grape juice was used as acid catalyst, self-made improved Soxhlet extractor was used as an extraction device. According to the experimental results, when the concentration of ethanol was 20%, the temperature of extraction tube was 60 ℃, the temperature of oil bath was 145 ℃, the dosage of grape juice was 10 mL, and the extraction time was 5 h, the total yield of ginsenoside Rg3 and Rg5 was 2.29%. Ginsenoside’s extraction and structure modification were compressed into one step, and solid-liquid separation could be done directly with less solvent due to the method, so that highly activity ginsenosides could be quickly and environmentally friendly prepared.
